Tempo-iSenso™ cells are human sensory neurons derived from human dermal cells, using human induced pluripotent stem cell (iPSC) lines under a fully defined proprietary induction and maturation conditions. Tempo’s iSenso™ cells are branched structures when plated as a monolayer in culture and express biomarkers such as beta-Tuj (neuronal specific), BRN3a and Peripherin. In addition, Tempo-iSenso™ cells express biomarkers such as ASIC, CNR1, NTRK, P2X4, SCN9a/Nav1.7, TRPA1, TRPV1, P2X3, and SCN10A/Nav1.8 (RT/qPCR data not shown).  iSenso™ can be used under maturation conditions or co-culture conditions (with Schwann cells or additional neuronal subtypes) for a variety of functional characterizations and drug candidate validation studies.

Sensory neurons belong to one of the most important neuronal subtypes in the human peripheral nervous system (PNS). In the PNS, these neurons are primarily responsible for conducting cations-mediated electrical activities and sensing external or internal stimuli (e.g., small molecules, peptides, etc). They form neuronal-glial networks that are responsible for a variety of motor and sensory mediated functions. They are implicated in several disabling neurological disorders, including pain, sensory neuron diseases, amyotropic lateral sclerosis (ALS) and mechano-or-temperature sensing deficiencies.

Technology used: an in-house developed proprietary feeder-free, serum-free, viral-free, genetic-vector-free, integration-free reprogramming method.

Specifications: ~1.0x10^6 cells per 1ml of freezing medium (vial)

Long-term Storage: liquid nitrogen

Growth Properties: adherent

Storage: remove cryovials (dry ice packaging) and place the vial into liquid nitrogen for storage. Alternatively, thaw and use the cells immediately.

QC: Sterility, Safety (BioSafety Level 2), HIV/viruses, bacteria, fungi: negative. Cell viability post-thawing (>90%)
